It depends on the relationship the sisters have and also of the sister you already made a move on's perspective of you. If you missed an EW or failed an escalation with her, its likely that she told her sister that you're "creepy" or "a player" depending on her deposition towards you. In other words, its basically "negative preselection". Having said that, if you're charming and charismatic enough, you can still win her over. And the bottom line is, there's no harm in trying. So I'd say, go for it.

She still would kiss me and flirt at work, but I stopped and it has stopped.
I want her sis now, they're kinda close I guess.

Was in a similar boat a couple of months ago. Didn't really know if I wanted the less cuter sister who was all over me. I dug her personality but wanted the hotter one. Long story short, the less cuter one won me over but it was too late, by then some other dude working there swooped her and is currently dating her. The hotter sister backed off before and after this... Guessing she was letting the younger one run her game on me without cockblocking....
Didn't kiss or get sexual with her though, mainly cuz I didn't want to be slotted in the b.f zone and I like keeping things business at work. She almost got to me though, indecision and uncertainty eventually won.
